Tricyclic antidepressants (TCAs)

Tricyclic antidepressants (TCAs) are an older group of drugs. They block reuptake of serotonin and norepinephrine, similar to SNRIs, but they’re messy in the sense that they act on several other targets, and as a result, they can have considerable side effects.

There are two main classes of TCAs based on chemical structure. Tertiary amines have a greater effect on serotonin, and secondary amines have more of an effect on norepinephrine. Amitriptyline (Elavil), imipramine (Tofranil), clomipramine (Anafranil), and doxepin (Silenor) are tertiary amines. Desipramine (Norpramin) and nortriptyline (Pamelor) are secondary amines. Desipramine is an active metabolite of imipramine, and nortriptyline is an active metabolite of amitriptyline.
